Drugs of abuse are small molecules that typically do not induce an antibody response following injection or inhalation. To induce antibodies against small molecules, structural surrogates of the molecules, which were named “haptens”, must be coupled to immunogenic proteins, called “carriers”. These structural surrogates are typically drug-linker adducts, in which the linker has a terminal functional group that forms a covalent bond with the carrier. The efficacy of these conjugate vaccines depends on several factors including hapten design, coupling strategy, hapten density, carrier protein selection, and vaccine adjuvant.
Our aim was to synthesize haptens of morphine and its derivatives. For this purpose we used the nor-molecules in addition reactions. These products are hapten precursors and they can be converted into the desired molecules, containing carboxyl or amine functional groups. After this step it is possible to couple them to amino acids or proteins.
As a result we developed three ways to obtain morphine derivative haptens. We synthesized over 40 previously unknown molecules. Their structures were confirmed with NMR and MS spectroscopy and the purity was checked with TLC.
At present there are several ongoing researches in this topic worldwide to obtain an effective vaccine against heroin, cocaine and nicotine. This fact gives grounds for our research as well.
