Some molecules are potentially important on tumor cell radio sensitivity while the combination of immunotherapy and these sensitizers could increase the therapeutic potential, there are no studies that analyze possible synergies between these. The pentoxifylline (PTF) a methylxanthine derivative). It has been showed that 0.5 to 1.0 mMOL of PTF elicited an increase in radiation-induced cytotoxicity, whereas the combination of this substance with radiation allowed an increase of the radiosensitization; the earlier studies showed that some alternatives for improvement the activation of NKT cells and survival of specimens with solid tumors exposed to radiotherapy is real. The alpha-galactosylceramide (KRN7000) is a glycosphingolipid able to inhibit tumor growth and to prolong the survival in treated mice. Study aim: To test the therapeutic potential of pentoxifylline in combination with alpha-galactosylceramide treatment and radiotherapy using a solid tumor model in mice. Method: The mice of each group (1:KRN7000, 2: PTF, 3: KRN7000+PTF, 4: control) was inoculated s. c. with 2x105 B16 cells and the tumor size was determinate, we take samples of blood for flow cytometer analysis and the tumor was recollected for immunohistochemistry analysis, the same groups was used for radiotherapy assay . Results: Activation of NKT cells was effective for tumor size reduction in combination with PTF, we can observed more antitumor effect than alone, we observed the survival of the mice was higher in the group with both compound and adding radiotherapy, the number of iNKT was higher in KRN7000 group than the others, and in morphological descriptions we observed positives changes in the KRN7000+PTF+Radiotherapy in comparison with the control groups. Conclusions: the results suggest the use of immunological therapy together with the administration of a radiosensitizing compound plus the main treatment shows morphological improvements in tumor tissue and reduces tumor size.
