Introduction: The etiology and pathogenesis of ulcerative colitis (UC), one of the inflammatory bowel diseases, are currently unknown. The colonic barrier disturbance plays an important role in UC development. Two components of this barrier are mucus and glycocalyx with their main structural components- highly glycosylated glycoproteins mucins.
Aims: to study colonic mucin barrier disturbances in mice with acute and chronic colitis induced by dextran sodium sulfate salt (DSS).
Methods: adult male mice C57Bl/6 were used (n=45). The mice received 1% DSS solution for 5 days and were sacrificed on day 7 (acute colitis) and 28 (chronic colitis). Animals of the control group received tap water. The medial part of colon was extracted. Histological sections stained with H&E, alcian blue (acidic mucins) and PAS-reaction (neutral mucins) were studied, as well as surface epithelium ultrastructure and the expression of mucus-forming Muc2 and glycocalyx-forming Muc1, Muc3 and Muc13 with qRT-PCR.
Results: In acute colitis there were ulcers, extensive neutrophilic and lymphocytic inflammatory infiltration, a goblet cells number reduction, an increase in their size and a decrease in the content of acid mucins. Ultrastructural examination revealed a large number of bacteria close to epithelial cells and a decrease in the glycocalyx thickness. Muc3 expression was reduced.
In chronic colitis there were epithelialized ulcers, extensive lymphocytic and plasma cells inflammatory infiltration. The number, size, acidic mucins content of goblet cells and glycocalyx thickness were unchanged, the number of bacteria in contact with epithelial was lower than in acute colitis. The expression of Muc1 and Muc2 was higher and Muc13- lower than in control group.
Conclusion: In acute colitis we found the disturbance of goblet cells, glycocalyx and mucus layer, decreased expression Muc3. In chronic colitis, there is a Muc1, Muc2 upregulation and a Muc13 downregulation.
