The change in the 5-HT content and the number of enteroendocrine cells in different parts of the colon in postnatal ontogenesis of rats.

Introduction: 5-HT plays a major role in the normal gut functioning both as a neurotransmitter and immunomodulator. 5-HT and the number of its main producers - enteroendocrine cells (EC-cells) change with age. However complex evaluation of both 5-HT and EC-cells changes in different regions of the colon and in different ages is lacking.
Aim: To investigate the age related changes of EC-cells populations and the 5-HT content in the homogenate of the proximal and distal colon and peripheral blood of rats different ages.
Methods: In our study 24 male Wistar rats of 4 different ages were used (n=6 in each group): newborn (2-4 days), prepubertal (25-30 days), adult (3 months) and aged (16-18 months). 5-HT was measured in the proximal and distal colon and peripheral blood. For EC-cells labeling the anti-chromogranin A antibodies were employed. The 5-HT content was measured with high-performance liquid chromatography.
Results: In the prepubertal rats, in comparison with the newborn, the number of EC cells increased 1.8-fold. The adult and aged animals had no differences. The 5-HT content in the colon homogenate progressively increased 15.8-fold from newborn to prepubertal animals, and 3.9 fold from prepubertal to the aged animals. The peripheral blood 5-HT content increased in adult animals and did not change in aged animals. In the newborn the number of EC-cells and the 5-HT content showed no difference between the proximal and distal colon. However, in the prepubertal, adult and aged animals these parameters were higher in the proximal colon in comparison with the distal colon.
Conclusion: During maturing and aging there is an increase in number of colon EC-cells and the colon and peripheral blood 5-HT content. In the proximal colon, in comparison with the distal colon, the number of EC cells and the 5-HT content is higher in all age groups, except for the newborn.
