Introduction: Myramyldipeptide (MDP), a monomer of prokaryotic peptydoglycan, is a potent immunostimulator. To resolve it’s drawbacks, multiple chemical modifications were performed. One of such modifications is the addition of aglycon. Glycosides of MDP are powerful immunomodulators which can be used as adjuvant drugs in oncology.
Aims: to investigate the effect of Heptyglycoside MDP on the growth and proliferation of B16 melanoma tumor in mice.
Methods: The tumor cells were implanted subcutaneously into the right side of back of adult C57BL/6 mice. The experimental mice were divided into two groups: the first group which had the Heptylglycoside MDP being administered sequentially during the whole course of the experiment; the second group which had the Heptyglycoside MDP been administered once before the tumor implantation. The control group consisted of animals with implanted tumor with administration of subcutaneous solution of NaCL 0.9% solution. Each of the experimental groups was further subdivided into three subgroups according to the dosage of Heptylglycoside MDP: 1mkg, 5mkg, 25mkg per animal (N=10).
The moribund condition was the end point and the measuring of the median survival time was performed. The volume of the tumor in each animal was measured on 10, 15 and 20th day after the implantation.
Results: None of the subgroups of the second group showed any statistical differences from the control group. The subgroups of the first group with the dosage of the Heptylglycoside MDP 5 and 25 mkg per animal showed significant tumor growth suppression on the 10th (57% and 74% respectively) and 20th days (40% for both groups) after tumor implantation.
Conclusion: The results suggest that the most effective tactics of use of the new MDP derivatives in oncology may be the constant adjuvant therapy in conjunction with chemotherapy.
