Morphological change of immune system organs in male C57Bl/6 mice with hyperandrogenemia

Introduction: Testosterone (T.) affects infectious and autoimmunity diseases via immune response and inflammation. Higher level of testosterone suppresses immune response. The majority of publications on T. immune effects focus on in vitro studies. Because anabolic-androgenic steroids are used both for sport performance and contraception there is an actually demand to study the relation between T. levels and inflammation.
Aim: To study morphological and immunological changes of testis and immune system organs in hyperandrogenemia.
Method: The oil-based solution of T. was injected intramuscularly three times to C57bl/6 mice with a carrier receiving control. We performed a morphological and morphometric analysis of immune system organs. Flowcytofluorometry methods were employed to axillar lymph node cells to measure the lymphocytic subpopulations.
Result: The serum level of T. significantly increased 12-fold in mice with hyperandrogenemia. In their testis seminiferous tubules were dilated with an increased luminal diameter. The volume of thymus lobes was reduced 3-5-fold. There was a devastation of cortex layer with an absence of clear margins between it and medullary layer and a partial layer inversion. Subcapsular zone was intact. In medulla there were epithelial thymic cells with sporiadic lymphocytes, whose number was reduced. Lymph nodes had normal structure with mostly secondary lymphoid nodules and large light centers, which contained lymphoblast and small amounts of lymphocytes and macrophage. Paracortical zone was densely populated by lymphoblasts. Medulla sinuses were enlarged with macrophages and lymphocytes in 1:1 ratio. Marginal sinuses were focally enlarged with small amounts of lymphocytes and macrophage. There was no significant difference in the relative number of CD3+CD4+Th-cells, CD3+CD8+cytotoxic and CD3-CD19+В-cells in axillar lymph nodes. Spleen hyperplasia was present with a white pulp area fraction 2.5-fold increase.
Conclusion: The male C57bl/6 mice with hyperandrogenemia showed an involution of thymus with a cortex and medulla devastation and a hyperplasia of lymph nodes and spleen white pulp.
