Atherosclerosis is a slowly progressing, pathological process involving the intima and media of large- and medium-sized arteries. Focal thickenings of the arterial wall or plaques gradually interfere with the flow during atherogenesis when thrombosis is superimposed on non-stenotic lesions, potentially leading to ischaemia of target organs and tissues.
The clinical importance of AB0 blood groups extends beyond the direct implications on transfusion and organ-transplantation compatibility. It is increasingly being recognized that individuals with non-0 blood groups may be at elevated risk of thromboembolic-, cerebrovascular ischemic events, myocardial infarction and peripheral vascular disease compared with individuals with blood group 0.
Endothelial dysfunction is an early and important trigger of atherogenesis, emerging as a response to a number of various oxidative, infectious and mechanical injuries. Von Willebrand factor (VWF) is a marker of these processes. VWF, a large multimeric glycoprotein that adheres to sub-endothelial collagen in order to catch bound platelets at high shear- and elongation forces and serves the platelet plug formation. It carries and protects coagulation FVIII that catalyze the coagulation cascade to form stable fibrin net.
Examination of 11,673 participants in the Atherosclerosis Risk in Communities (ARIC, www.cscc.unc.edu/aric/) prospective epidemiologic study demonstrated the intrinsic effects of ABO on VWF and factor VIII, independently.
Osteoprotegerin (OPG) is a dimeric glycoprotein with a multifunctional cytokine role and highly expressed in a large number of tissues. They are associated in plasma, co-localized in, and co-secreted from the Weibel-Palade bodies of the endothelial cells.
Our group evidenced that AB0 blood group type is determinant of OPG levels in blood, similar to its role on VWF.
Next we search for co-localization of the above three molecules from vessels of patients with different stage of the atherogenesis to better know the pathophysiological role of them.
