Morphological and immunological features of chronic ulcerative colitis in mice with different resistance to hypoxia

Introduction
Hypoxia contributes to development of inflammation, because signal pathway, activated by hypoxic action (HIF-1), is associated with NF-kB. However, there are no data of features of inflammation in mice with different resistance to hypoxia.
Aims
To characterize morphological and immunological features of inflammation in chronic colitis induced by dextran sodium sulfate (DSS), in mice with different resistance to hypoxia.
Method
The resistance of male C57Bl/6 mice (n=60) to hypoxia was determined in hypobaric chamber by measuring the time taken for onset of gasping at critical altitude of 10000 m. Tolerant animals had GT more 10 minutes, susceptible – less than 3 minutes. One month after the hypobaric test, chronic colitis was induced using 1% DSS and subpopulation composition of peripheral blood lymphocytes was determined.
Results
Differences in the intensity of DSS-induced inflammation in colon of mice with different resistance to hypoxia were revealed. The relative prevalence of ulcers with epithelialization in the mucosa in susceptible mice was 21.5 (19.9-23.4)% - significantly higher (p = 0.02) than in tolerant animals 14.9 (13, 0-16.2)%. The relative length of ulcers without epithelization in the compared groups did not differ. The susceptible mice showed during chronic ulcerative colitis a decrease in absolute and relative amounts of T-lymphocytes, T-helper cells, cytotoxic T-lymphocytes, T-regulatory cells, and an increase in relative number of B-lymphocytes. In peripheral blood of tolerant mice only relative number of T-lymphocytes, an absolute and relative number of T-helpers and T-regulatory cells decreased. Compared with susceptible animals, tolerant mice had a greater absolute amount of cytotoxic T-lymphocytes, as well as the absolute and relative number of T-regulatory cells.
Conclusion
Thus, in peripheral blood of hypoxia susceptible mice with chronic colitis, content of T-lymphocytes, T-helpers, cytotoxic T-lymphocytes, T-regulatory cells decreases, but the number of B-lymphocytes increases. Compared with tolerant mice, prevalence of ulcers with epithelization in colon was higher in susceptible animals.
