Cirsium erisithales (Jacq.) Scop (Asteraceae family) grows in European mountains, in the 600-1600 m height. Its achene fruits accumulate valuable secondary metabolites called lignans. Lignans are phenoloid type molecules, showing biological activities like antioxidant, antiinflammatory and antiviral effects. They are present in glycoside or aglycone form in the fruit, together with specific endogenous enzymes with the ability to cleave the glycosidic bonds.
The aim of our study was to characterize the secondary metabolite content of the fruit.
Two methanolic extracts were made: one directly from the powdered, intact fruit sample and another after endogenous enzymatic treatment. LC-ESI-MS/MS and NMR techniques were used for structure elucidation of the compounds. Quantitative measurements and isolations were performed by HPLC-UV and preparative HPLC, respectively. Antitumor effect was investigated on SW480 colon adenocarcinoma cell-line, with SRB colorimetric assay.
Lignan glycosides present in Cirsium species can be easily hydrolysed by specific plant endogenous enzymes. In order to determine the fruit composition of C. erisithales for the first time, composition of intact (untreated) fruit extracts as well as their endogenous enzyme-treated correspondents were compared. Two lignan glycosides (arctiin, tracheloside) and three aglycons (arctigenin, trachelogenin and norbalanophonin) showing dose-dependent antiproliferative activity were determined.
