Introduction:
Lymphatic vessels are important for the maintenance of healthy organs. Changes in lymphatic function or morphology are known to play a role in the development of lymphedema, atherosclerosis, high-blood pressure and other metabolic diseases. The pathophysiological role of lymphatic vessels is not fully understood yet, mainly due to the lack of visualization techniques.
Aim:
In this study we aimed to develop a new technique to visualize organ-specific lymphatic growth.
Materials and Methods:
To visualize organ-specific growth of lymphatics, different organs of CL57BL/6 and Prox-1GFP reporter mice were tissue cleared, followed by whole mount immunostainings. In parallel, we performed paraffin based histology, followed by immunostainings to show the difference between traditional visualization methods and our newly developed visualization technique. Flt4+/+ and Flt4kd/+ mice were used as a model for lymphatic alterations. Pictures were taken by upright, stereo and confocal microscopy.
Results:
We could demonstrate that tissue clearing, followed by whole mount immunostaining, is a very effective way to visualize organ-specific lymphatic growth, especially in comparison to older techniques such as simple immunostaining of paraffin sections. With this method we are able to visualize lymphatic vessels in 3D, show differences in the lymphatic morphology between organs and compare and quantify the lymphatics of different mouse models.
Conclusion:
Our results show that tissue clearing in combination with whole mount immunostaining is a great technique to visualize the organ-specific growth of lymphatic vessels. This new approach can help to clarify the role of lymphatic vessels in the pathology of various diseases.
