A Comparative Study on the Effect of Different Oils on the Properties of Poloxamer-Based Hydrogels for Vaginal Candidiasis Treatment

Vaginal candidiasis is a common infection among women worldwide, and the available treatments are often associated with adverse effects. Hydrogels and emulgels have been proposed as potential vehicles for drug delivery due to their biocompatibility and ability to enhance the stability and bioavailability of the active ingredients. In this study, we aimed to develop and evaluate emulgels based on poloxamer 407 and 188, with several types of oil and lactic acid, for the treatment of vaginal candidiasis.
We prepared different hydrogels using poloxamer 407 and 188 in various percentages and evaluated their gelling temperature using a Malvern Kinexus Pro rheometer. From the hydrogels, we prepared emulgels using different oils (almond, Medium-chain triglycerides (MCT), sunflower, jojoba, oleic acid) and observed their effect on the gelling temperature. To ensure the antifungal/antibacterial properties and to create the pH necessary for the growth of lactobacilli of the emulgels, we added lactic acid and adjusted the pH between 3.6-4.2. We evaluated the effect of lactic acid on the gelling temperature and monitored the stability of the emulgels by measuring the drop sizes for five days.
Our results showed that the gelling temperature of the hydrogels varied with the percentage of poloxamer 407 and 188, with higher percentages resulting in lower gelling temperatures. The addition of oils had a significant effect on the gelling temperature of the emulgels. Emulgels made with of MCT oil, almond oil and oleic acid remained unchanged in viscosity for the longest time. The emulgels were stable for five days, with no significant changes in drop sizes, but viscosity was observed to increas after two weeks in most of the emulgels. The addition of lactic acid lowered the viscosity of the emulgels and increased the gelling temperature. The pH adjustment resulted in a pH suitable for vaginal administration.
Our study demonstrates the potential of poloxamer-based emulgels as vehicles for the treatment of vaginal candidiasis. The addition of lactic acid and pH adjustment could enhance the antifungal properties of the emulgels. Further studies are needed to evaluate the efficacy of the emulgels with different chosen active ingredients.
