Introduction
The importance of the serotonin neurotransmitter system in aggressive and impulsive behaviors is well documented. The serotonin transporter and serotonin autoreceptors (HTR1A and 1B) play a crucial part in determining the synaptic serotonin level. Some of their genetic variants have been shown to affect gene expression, e.g. the promoter region of the HTR1A gene contains a functional polymorphism (-1018C/G, rs6295), whereas the rs13212041 polymorphism in the non-coding region of the HTR1B gene was shown to alter miR-96 binding and its low expression variant was associated with greater anger and hostility.
Aim
The aim of the current study was to assess the association between aggressive/delinquent behaviors and polymorphisms regulating serotonin expression.
Method
315 adolescent subjects from a community group (50.6% male, mean age:15.9 years [SD:1.06 years]) and a clinical ADHD group (73.7% male, mean age:15.6 years [SD:1.1 years]) were carefully phenotyped on a spectrum of impulsive, delinquent and aggressive behaviors through multiple informants: self-, parent- and teacher report questionnaires (Buss-Perry Aggression Questionnaire, Self-Report Delinquency Scale, Youth Self-Report, Disruptive Behavior Disorder Rating Scale). Genotyping was also performed on polymorphisms that affect serotonin levels. HTTLPR serotonin transporter gene (20-23 bp VNTR region), serotonin autoreceptors 1A (rs6295), 1B(rs130058), 2A(rs6311), and the MAOA gene (5’ region 30bp VNTR). Associations between polymorphisms and behavior were assessed separately in the community and clinical subsample.
Results
The most consistent association results showed with aggression and delinquent behavior.
Conclusion
Differences across with and without ADHD groups with regard to associations between genetic polymorphisms and aggressive / delinquent behavior indicate that certain genetic variants may differentially confer risk for affective outcomes, given ADHD status. That findings held across multiple informants suggest results may be robust.
