Historically, extracts from the bark stem of Cinchona officinalis containing quinine and alkaloids have been utilized to treat fever and malaria. Recent research has uncovered apoptotic properties in cancer cells attributed to derivatives of cinchona alkaloids. Likewise, chalcones, plant-based polyphenolic compounds, are known for their anti-fungal, antimicrobial, antimalarial, and anticarcinogenic properties. In this study, we defined the cytotoxic and antiproliferative effects of synthetic hybrids combining cinchona and chalcone in human cancer cell lines. Specifically, two derivatives, AD-12 and AD-13, exhibited IC50 values below 2 µM in lung squamous cell carcinoma (EBC-1) cells. Our findings demonstrate that AD-12 and AD-13 increased intracellular ROS levels and induced caspase-3/7 and -8 activity in EBC-1 cells, leading to apoptosis. Furthermore, these apoptotic effects coincided with a temporary inhibition of P-gp efflux activity, while the expression levels of P-gp transporters remained constant. Collectively, our study highlights the promise of cinchona-chalcone hybrids as potential candidates for novel anticancer therapeutics.
