Introduction - Callous-unemotional (CU) traits, characterized by shallow affect, lack of empathy, and fearlessness, are associated with ADHD and related disorders like ODD, CD, and ASPD. The serotonergic system, involving serotonin and related genes (e.g., SLC6A4, HTR1A), plays a key role in mood regulation and behavior. Research indicates that polymorphisms like 5-HTTLPR and HTR1B are linked to ADHD and CU traits, with varying effects based on genetic variations. Existing studies suggest that CU traits mediate the association between 5-HTTLPR and ADHD, highlighting the need for further investigation.
Aims - Aim was to examine whether accounting for self-reported negative affectivity (NA) and parent-reported oppositional defiant disorder (ODD) symptoms as relevant covariates, parent-reported ADHD risk moderates the association between serotonergic polymorphisms and prospective changes in self-reported CU traits in adolescents. Multi-locus genetic profile (MLGP) scores captured the combined effect of included polymorphisms.
Methods - Data from a larger longitudinal study were analyzed. Participants (243 adolescents) underwent cognitive, clinical, genetic, and questionnaire assessments. ADHD risk was determined using the parent-rated ADHD Rating Scale-5, with classifications based on symptom criteria. Genotyping for serotonergic polymorphisms (HTTLPR, HTR1B) was performed.
Results - The robust regression model with 5HTTLPR polymorphism predicted: ΔCallousness scores, with a positive association of ΔUncaring and ADHD risk by 5HTTLPR polymorphism interaction with ΔCallousness scores; ΔUncaring scores, with a positive association of ΔCallousness with ΔUncaring scores. The robust regression model with the serotonergic MLGP score predicted: ΔCallousness scores, with a positive association of ΔUncaring and negative association of the ADHD risk by serotonergic MLGP score interaction with ΔCallousness scores; ΔUncaring scores, with a positive association of ΔCallousness with ΔUncaring scores.
Conclusion – Serotonergic polymorphisms differentially confer risk on CU traits based on ADHDrisk status.
