Introduction
ARHGAP25 is a Rac-specific GTPase activating protein playing a key role in immunological processes. Its function in neutrophilic granulocytes has been thoroughly elucidated, followed by the discovery of its importance in B lymphocyte biology. However, our current understanding of its function in T-cell biology is still limited.
Aims
We aim to investigate the role of ARHGAP25 on T cell maturation and functions, searching for novel ways to combat inflammatory and autoimmune diseases.
Method
We used healthy adult Arhgap25+/+ (wild type, WT) and Arhgap25-/- (knock-out, KO) mouse strains with C57BL/6 background, comparing T cell levels in peripheral blood, along with spleen, inguinal and axillary lymph node and thymus sizes and cellular compositions. T cells isolated from healthy spleens were cultured in RPMI-1640 medium to assess their viability and cellular functions.
Results
Peripheral blood of KO mice showed decreased CD3+ T cells with no change in CD4/CD8 ratios. Similar differences were detected in peripheral lymph nodes, resulting in a smaller size. The spleen showed no difference, but the thymus was significantly enlarged, with higher thymocyte counts and altered subpopulation ratios.
Conclusion
Apart from being a key player in innate immune functions, ARHGAP25 acts as a regulatory molecule in T cell development. The lack of ARHGAP25 protein disturbs healthy T-cell maturation and causes unusual thymus enlargement with a decreased T-cell count in the blood.
