Aging is a multifaceted process that involves a decline in functionality and integrity. Among others, it triggers the development of a chronic inflammatory state and gut dysbiosis1. rag1 mutant zebrafish has been reported to develop accelerated aging characterized by increased expression of inflammatory and apoptotic markers and systemic macrophage infiltration, likely due to over-activation of innate immunity 3. Aiming to deepen the potential roles of microbiota in inflammatory aging, rag1 mutant zebrafish was used to perform a variety of omic technologies together with histological and enzymatic analyses. The results showed a reduced villi length and goblet cell number in the intestine of rag1 mutants together with several changes in genes related to lipid metabolism (slc5a8l), eicosanoid production (lta4h, ptges) and apoptosis (casp6a, casp6b.1), which correlated with higher caspase-6 activity. Alongside with this, a reduced bacterial diversity, increased Proteobacteria and decreased Fusobacteria abundance, together with a reduction in genes related to short-chain fatty acid metabolism was observed in the gut microbiota of rag1 mutants. The evidences presented in this study show that in the absence of lymphocytes, the gut microbiota and intestinal homeostasis undergo significant and detrimental changes. These findings shed light on the critical interplay between the gut microbiota and host immunity, offering novel therapeutic approaches for age-related diseases.
