Introduction. Myocarditis is a polyetiological inflammatory heart disease with a wide range of clinical manifestations, from asymptomatic to life-threatening conditions such as heart failure and sudden cardiac death.

Aims. The aim of this work was to investigate the immune response and molecular changes in the myocardium of male BALB/c mice at 14, 28, 42 and 60 days after immunization.

Methods. Myocarditis was modeled in adult male (n=20) BALB/c mice, which got subcutaneous injection of 150 μg of α-myosin (Med Chem Express) with 120 μL of complete and incomplete Freund's adjuvant (Sigma) on days 0 and 7. Control group with Freund's Adjuvant injection (n=10) was injected with 120 μL of complete and incomplete Freund's adjuvant (Sigma) on days 0 and 7. Control group animals males (n=10) were not injected. Animals were sacrificed 14, 28, 42 and 60 days after the second immunization by decapitation. The level of Hif1α, Nfκb, Tgfβ, Il1, Il6, Il10 expression in the myocardium was assessed by RT-PCR. The number of immune cells and fibroblasts in the myocardium was determined on histological slides stained with H&E. The data were processed in «Statistica» (Kruskall-Wallis test, Dunn’s multiply comparisons test).

Result. The number of immune cells and fibroblasts in the myocardium was higher in the experimental groups than in the control groups, which can indicate the development of myocarditis. There was no difference in gene expression between the control group and the group with the injection of Freund's adjutant. Compared to the control group, on 14 days the expression of Nfκb and Il1 increases and then decreases sharply. Il10 and Il6 expression increases at 42 and 60 days. Expression of Hif1α and Tgfβ increases on days 14 and 42.

Conclusion. Changes in the dynamics of gene expression have been revealed. Markers of acute inflammation were higher at 14 days, while markers of chronic inflammation and anti-inflammation activity were higher at late stages. The first increase of Hif1α is associated with the inflammation, and at 42 days it is associated with the adaptation response. The increase of Tgfβ may be associated with fibrosis. According to the data can be observed in the development of myocarditis, should be investigated physiological changes during myocarditis.
