Function of the median raphe in the regulation of hippocampus

Introduction:
The median raphe region (MRR) is an important mood regulatory center of the brainstem. However, the function of the MRR vesicular glutamate transporter 3 (vGluT3) positive cells is poorly understood. Since their activity is enhanced by negative experiences, we hypothesized that their projection pathways contribute to the processing of negative experiences.
Objective:
In this work, we aim to map these pathways and understand their functions.
Methods:
We mapped the target-selective connections of MRR vGluT3 neurons using adeno-associated viral (AAV) fluorescent pathway tracing, and investigated their function using AAVs encoding photosensitive opsins and optogenetic methods. Histological processing was complemented by immunohistochemical techniques.
Results:
Our results showed that MRR vGluT3 neurons in the stratum lacunosum of the cornu ammonis regions of HIPP and in the hilus of the gyrus dentatus mainly innervate interneurons containing neuropeptide Y and calbindin. Our behavioural experiments suggest that these cells may play a role in linking events that do not overlap in time.
Conclusions:
Our results contribute to our understanding of the role of brainstem networks in the processing of negative experiences, which may also help to identify potential therapeutic targets.
