Introduction: The mechanism of the development and progression of cardiovascular diseases, such as atherosclerosis, is still not fully understood, which is a major health challenge worldwide. The disease develops due to the accumulation of lipid plaques on the walls of blood vessels, leading to thickening and narrowing of the vascular walls. Macrophages have been described as crucial mediators of the atherosclerotic cascade; however, their role has not been fully elucidated.
Aims: Our aim is to improve our understanding of the pathogenesis of atherosclerosis and to develop a new therapeutic option for the treatment of the disease. We aim to achieve these goals through a detailed preclinical study of an experimental agent, ACHA, developed by our collaboration partner, AnnaHorist Ltd.
Methods: We used an immortalized macrophage cell line and tested whether these cells take up the ACHA. We employed the transgenic LDLR -/- mouse model to induce atherosclerosis through a 16-week high-fat diet. Following disease induction, mice received intravenous administration of the ACHA compound. Blood samples were collected to measure cholesterol, LDL, and HDL levels, aortic arches were analyzed for lipid accumulation.
Result: Our findings indicate that macrophages take up the ACHA as early as 5 minutes. Furthermore, our results showed a sex difference in disease progression, more lipid deposition was observed in female mice than in males. Stereomicroscopic images revealed a decrease in aortic plaque formation, suggesting the potential therapeutic effects of ACHA; however, further investigations are needed to optimize its efficacy.
Conclusion: These results suggest that ACHA may have beneficial effects in moderating atherosclerosis progression, although further studies are required to optimize dosage and administration protocols. We would also like to investigate the difference in disease progression between sexes. Our findings contribute to the ongoing exploration of novel treatment strategies for cardiovascular diseases, highlighting the importance of targeted molecular interventions.
