Primordial germ cells (PGCs) are the stem cells of the germline, playing a crucial role in gametogenesis and the transmission of genetic information. Accurate identification and characterization of PGCs are critical for research in developmental biology, germline preservation, and reproductive science. This research project aims to characterize a novel mouse monoclonal antibody (clone name: 30B6) originally generated by our research group for the detection of B-cells. Preliminary immunocytochemical analyses revealed that, in addition to labelling early B-cells in the bursa of Fabricius of avian embryos, 30B6 also specifically recognizes PGCs. In avian species, reliable identification of PGCs requires multiple antibodies, as no single marker uniformly labels PGC populations across developmental stages. We hypothesize that 30B6 exhibits greater sensitivity and specificity for PGC detection than currently available markers and may have broader applicability across various species and developmental stages. We examined the binding characteristics and specificity of the antibody in different developmental stages of chicken embryos and their lymphoid organs using immunohistochemical and immunofluorescent staining techniques. Our findings indicate that the 30B6 antibody specifically labels PGCs in birds from early developmental stages, showing intense immunoreactivity in the germinal crescent of one-day-old embryos. 30B6 offers more distinct labelling of PGCs in the gonadal ridges compared to commercially available markers. Additionally, 30B6 recognizes goblet cells and their product lining the intestinal epithelium in birds (chicken, quail, partridge), mammals (human, not rodents), and reptiles, suggesting broader cross-species specificity. Furthermore, 30B6 stains the human primordial germ cells in an 8-week-old embryo. Taken together, these results demonstrate that the 30B6 monoclonal antibody is a novel and versatile tool that can enhance the accurate identification of avian and human PGCs and support further research in germline development.
