Introduction: Isomalt is a widely used excipient in the food and pharmaceutical industries. It consists of two sugar alcohols: 6-O-α-D-glucopyranosyl-D-sorbitol (1,6-GPS) and 1-O-α-D-glucopyranosyl-D-mannitol dihydrate (1,1-GPM). Among its advantageous properties, its thermal stability is particularly noteworthy, as it enables its application in the production of medicated lollipops. Such dosage forms offer several benefits, including a rapid onset of local therapeutic action and suitability for patients with swallowing difficulties.
Aims: This study aimed to develop medicated lollipops using compression and molding techniques, employing two types of isomalt: type 720 (1:1 GPS:GPM) and type 721 (3:1 GPS:GPM). Furthermore, the study aimed to compare and evaluate lollipops prepared by the different manufacturing methods and formulated with different types of isomalt, as well as to investigate the incorporation of lidocaine hydrochloride as a model active pharmaceutical ingredient (API).
Method: For the preparation of compressed lollipops, sodium stearyl fumarate was used as an excipient at a concentration of 1% w/w. In the case of molded lollipops, melted isomalt was poured into silicone molds. Various quality assessments were performed, including mass uniformity, breaking strength, disintegration, and dissolution testing. In addition, content uniformity of lidocaine hydrochloride was determined spectrophotometrically.
Results: Medicated lollipops were successfully prepared using two types of isomalt and two different manufacturing methods. The content uniformity of lidocaine hydrochloride was successfully determined in both compressed and molded formulations, and dissolution testing was performed.
Conclusion: The experimental results demonstrated that isomalt is suitable for the preparation of medicated lollipops using both compression and molding techniques. Furthermore, lollipops containing lidocaine hydrochloride were successfully produced.
