During aging, nicotinamide adenine dinucleotide (NAD+) depletion and impairment of cardiac ischemia tolerance can develop.
Our aim was to explore whether treatment with NAD+ precursor (nicotinamide mononucleotide, NMN) improves heart function and prevents enhanced ischemic damage in old hearts.
After pretreatment of young and old (4 and 26 months old) C57Bl/6N male mice with NMN (500 mg/kg/day ip.) or carrier (PBS) for 2 weeks, echocardiographic examination was performed. Afterwards, hearts were isolated and perfused with constant pressure in a Langendorff system. The hearts were exposed to 23 minutes of global ischemia, followed by a 120-minute reperfusion period. Coronary flow and left ventricular (LV) pressure changes were measured, and LV infarct size was determined.
In elderly hearts, echocardiography showed enlargement of the left ventricle, however ejection fraction was preserved. NMN treatment did not affect the echocardiographic parameters. The ratio of the infarcted area in LV of old mice was significantly higher compared to the young mice. After NMN pretreatment, the myocardial infarction size of the old hearts decreased. The postischemic coronary flow and functional restitution of old hearts were impaired compared to young controls, however NMN treatment restored this impairment.
Pretreatment with NAD+ precursor does not affect the resting heart function, whereas it significantly reduces the consequences of cardiac ischemia in hearts of aged mice; it has a favorable effect both on infarct size and on postischemic functional recovery.
